No more free rides for 'piggy-backing' viruses

ScienceDaily (Jan. 4, 2012) ? Scientists have determined the structure of the enzyme endomannosidase, significantly advancing our understanding of how a group of devastating human viruses including HIV and Hepatitis C hijack human enzymes to reproduce and cause disease.

The findings open the door to the development of new drugs to combat these deadly viruses that infect more than 180 million people worldwide.

The team of international scientists led by and Professor Gideon Davies from the University of York and Associate Professor Spencer Williams from the University of Melbourne, studied bacterial endomannosidase as a model for the same human enzyme and successfully determined the three dimensional structure of the enzyme using state of the art synchrotron technology.

Professor Davies, of the Department of Chemistry at York, said that knowing the structure of the enzyme revealed details on how viruses play biological "piggy-back," borrowing our cellular machinery to replicate and cause disease.

"If we understand how the viruses use our enzymes, we can develop inhibitors that block the pathway they require, opening the door to drug developments," he said.

In the past the problem has been that this group of viruses including HIV, Hepatitis C, Dengue Fever and West Nile virus, are able to bypass the main pathway if inhibited and replicate via a second pathway using this enzyme. Thus for a treatment to be effective, both pathways need to be blocked.

"It was already known how to block the main pathway for these viruses but until now, this endomannosidase bypass pathway has proved a considerable challenge to study," Professor Davies said.

Dr Williams said: "Combining international resources and expertise, we were able to determine the endomannosidase structure and this has revealed how we can block the bypass route, stopping the viruses from hijacking human enzymes."

Professor Davies added: "We hope that the work will lead beyond viruses and will point the way towards similar treatments for other diseases including cancer."

The research received funding support from the Biotechnology and Biosciences Research Council and the Australian Research Council.

The study is published in the Proceedings of the National Academy of Sciences (PNAS) this week.

Carl Peterson involved in interviewing Fisher as coach

Carl Peterson, the former Kansas City football czar and long-time friend of Dolphins owner Steve Ross, is involved in the interviewing of the new Dolphins head coach, two sources said on Tuesday.

Peterson stepped off the helicopter that landed at Dolphins headquarters in Davie with Ross and candidate Jeff Fisher, the sources said.

It's not known whether this means Peterson will have an active role in the franchise or if he's simply doing this as a favor for Ross and general manager Jeff Ireland. On Monday, Ireland announced he and Ross were formulating their plan to carry out the interviews.

Ross said a few weeks ago that he hadn't discussed a role with Peterson and that Ireland was his general manager.

The Dolphins announced just after 3 p.m. the interview with Fisher was completed.

Belarus limits use of 'foreign' websites, while Kuala Lumpur mandates WiFi in eateries

Talk about polar opposites. In an update posted to the Library of Congress, we're told that the Republic of Belarus will begin fining citizens that host domestic sites on "foreign" domains. Crazy? Definitely, but no less true. Starting later this week, any Belarusian not registered as an entrepreneur may use "only domestic internet domains for providing online services, conducting sales, or exchanging email messages." According to the interpretation, it "appears that business requests from Belarus cannot be served over the internet if the service provider is using online services located outside of the country," and police (as well as the secret police) are authorized to "initiate, investigate, and prosecute such violations." Wilder still, owners of internet cafes could have their entire business shut down if users are found to be accessing external sites on those networks, and for those curious, the law "may" extend to browsing within one's private home.

In a land a bit closer to the equator, it seems as if officials have their heads in a far more sensible place. Kuala Lumpur -- already home to one of the world's most lust-worthy airports and some of the most accessible / affordable mobile data plans -- will soon mandate that all new restaurants provide WiFi to their customers. And by "WiFi," we mean "access to the entire internet." According to the New Strait Times, the rule will be enforced as early as April, applying to eatery owners operating on premises larger than 120 square meters. We're told that existing owners will be forced to comply when renewing their license, and while the waves won't have to be given away for free, they'll be encouraged to charge no more than a "reasonable fee." Furthermore, the government is considering dipping into its own pockets in order to extend gratis WiFi to public facilities in the city, likely as a follow-up plan to the expiring WirelessKL contract. As if going to wander through Batu Caves and the colorful streets shown above weren't enough reason to visit Malaysia...

SEOUL, South Korea (AP) ? South Korea's president opened the door Monday to possible nuclear talks with North Korea and warned the neighboring country to avoid any provocations, saying the Korean peninsula is at a crucial turning point.

Lee Myung-bak's comments in a nationally televised speech come as the young son of the late Kim Jong Il takes power in North Korea as Supreme Commander of the military and ruling party leader after Kim's death last month. The North vowed Sunday in a New Year's message that it would bolster its military and defend the son, Kim Jong Un, "unto death."

"The situation on the Korean peninsula is now entering a new turning point," Lee said. "But there should be a new opportunity amid changes and uncertainty."

Lee warned that South Korea would sternly respond to any North Korean provocation. In 2010, 50 South Koreans died in attacks blamed on the North, though the Koreas have met in recent months for preliminary nuclear discussions.

North Korea, which has tested two atomic devices since 2006, has said it wants to return to long-stalled six-nation talks on halting its nuclear weapons program in return for aid. Washington and Seoul, however, have insisted that the North first show progress on past disarmament commitments.

If North Korea halts its continuing nuclear activities, Lee said in his speech, negotiations could resume.

"We are ready to resolve security concerns on the Korean peninsula and provide assistance to revive North Korea's economy through agreements in the six-nation talks," Lee said.

North Korea, however, has so far taken a hard line toward the South. It warned last week that there would be no softening of its position toward South Korea's government after Kim Jong Il's death. North Korea's powerful National Defense Commission said the country would never deal with Lee.

On Monday, Pyongyang accused Lee of plotting to shake up the North by temporarily placing his troops on high alert after Kim Jong Il's death. The North's main Rodong Sinmun newspaper said in a commentary carried by the official Korean Central News Agency that Lee must "kneel down and apologize" for his acts.

North Korea has regularly criticized Lee since he took office in 2008 and ended a no-strings-attached aid policy toward the North. Lee sought to link aid to progress in North Korea's nuclear disarmament.

The six-nation nuclear talks involve the two Koreas, the United States, China, Russia and Japan.

The Korean peninsula remains technically in a state of conflict because the 1950-53 Korean War ended in a truce, not a peace treaty.

North Korea's New Year's message, however, didn't include the country's routine harsh criticism of the United States and avoided the country's nuclear ambitions, a suggestion that the North may be willing to continue talks with Washington to win food aid.

This year is a crucial one for North Korea as it tries to build a "great, prosperous and powerful nation" befitting the April 2012 centenary of the birth of national founder Kim Il Sung, the new leader's grandfather.

In the North Korean capital, Pyongyang, New Year's Day was cold, snowy and gray, with splashes of bright color provided by flags, lanterns and banners reading "Happy New Year" that decorated the streets. Coming just days after the official mourning period for Kim Jong Il ended, the celebration was muted, unlike past years when cheerful people in their best holiday clothes thronged the streets.

Kim Jong Un, meanwhile, spent part of the day visiting a tank division, state media said, in what was apparently his first reported military field inspection since his father's death. Kim Jong Il regularly visited military units, factories and farms across the country, and his son's trip provided further evidence of the North's intention to link him closely with the military.

India, Australia bank on similar tactics

SYDNEY: They say change is the only constant, but in the cocoon of Indo-Oz cricket it pays to be static. Their teams may be at different ends of the spectrum, but rival skippers MS Dhoni and Michael Clarke have their own compulsions for sticking to the same XI and similar tactics - for the second Test at the SCG - that had been employed at the MCG last week.

Clarke, who was toying with the idea of fielding an all-pace attack on his home ground - which is hosting its 100th Test - quickly changed his stance and named an unchanged side on Monday after taking one look at the pitch that, according to curator Tom Parker, should aid spinners towards the later stages of the match.

This means pacer Ryan Harris will be the 12th man. Clarke argued that the Australian team management found it impossible to ignore the effort by the players who starred in the Melbourne win. "It's a tough call on 'Rhino' (Harris), he's been such a wonderful bowler for us and it was tough to leave him out," Clarke said. "The success we had and the way we performed in Melbourne, it was tough to change that team," he added.

Dhoni, who loves to sleep over team matters, kept the decision pending, but dropped enough hints that India, too, would go in with an unchanged side. Dhoni said, "In the absence of a full-fledged all-rounder in the side, playing a fifth bowler is not feasible."

Asked if he would like to give his three pacers Ishant Sharma, Zaheer Khan and Umesh Yadav the first use of the greenish pitch, Dhoni kept his cards close to the chest.

"It will depend on exactly how the wicket looks before the start of the game... how much moisture is in the wicket and how hot or cloudy the conditions are," he said.

Clarke, who feels winning the toss and batting is a Standard Operational Procedure (SOP) in Test matches, is likely to walk the talk as Australia may not want to risk batting last. Australia's new coach Mickey Arthur has backed lone off-spinner Nathan Lyon to cope with an Indian onslaught at the SCG.

"There's no doubt that they will try and go after him, but if we have runs on the board, Nathan becomes a very interesting prospect for us," Arthur said.

The accent, however, will be on pace in the 'Battle for Sydney' as the SCG waits for new reputations to be made and a few old ones to tumble. The one person on the threshold of history seemed least affected by it. For Sachin Tendulkar, it was just another day in office as he batted and batted in the nets against hundreds of 'throw-downs' from 18 yards. Tendulkar, who has not put a foot wrong at his favourite overseas venue since making his maiden trip to Australia in 1992, will be focused on getting a big score here that would give him a century of centuries and a beleaguered Team India a shot in the arm.

India badly need runs from its most consistent performer over two decades. Skipper Dhoni had been quick to blame the 'Melbourne Malady' on batting failure. India cannot afford another anaemic display from their stalwarts as well as their captain.

The same matrix applies to Australian batsmen, particularly to senior pros Ricky Ponting and Michael Hussey, who are fast reaching their sell-by date. Ponting, who made 62 and 60 at the MCG - the exact number of runs (122) Australia won by - will be looking to get a big one, for the road, at his favourite venue where he has so far amassed 1346 runs at 64.08. Ditto for Hussey who, like Ponting, finds himself listed on Cricket Australia's perform-and-play scheme.
